IGF-1 LR3 (Long R3 Insulin-like Growth Factor 1) is a synthetic analog of IGF-1 modified with an arginine substitution and an extended N-terminal peptide. Those modifications reduce its binding to IGF binding proteins (IGFBPs), which extends its active half-life from about 15 minutes (native IGF-1) to closer to 20-30 hours. IGF-1 is the downstream effector of growth hormone — LR3 skips the GH-to-IGF conversion step and delivers the anabolic signal directly.

Binds IGF-1 receptors on muscle cells and drives PI3K/Akt/mTOR signaling — the primary anabolic pathway for protein synthesis. Also stimulates satellite cell proliferation and differentiation, expanding the pool of nuclei available to muscle fibers. Partial insulin-receptor cross-reactivity means it also drives glucose uptake.
